Thumbs up for Orianthi from me as well - saw her play with Alice Cooper a couple of years ago and she was sublime. Only my opinion, but better than Nita above who replaced her in Alice's band.

Nancy Wilson from Heart has been doing great things for decades, she might have been overshadowed by Howard Leese in the early days but is a very capable guitarist in her own right.
I have tried to listen with my ears and not my eyes, and I'll happily go with Megi's selection, and Memphis Minnie from Bry's post - I'm way too old for heavy rock, metal and shredding regardless of how good the player looks!
About as far as I can comfortably go on the rock front in the context of this thread would be Joanne Shaw Taylor. Light and shade, playing with emotional depth, wide dynamic range and the guitar serving the song, rather than just being a display of technical skill. I do like the vocal style too, which helps.
An example below - sadly my favourite part of the solo is from 3:27 to 4:25, but the contrast with the more aggressive part of the solo that follows works well - and I can hear references to the melody line running through the faster part too - not just random shredding.
